Laboratory of Human Microbiom and Means of Its Correction

The scientific direction of endoecology, the founder of which was academician of the Academy of Medical Sciences of the USSR I.N. Blokhina, was formed at the institute in the 60-70s of the last century. Human microbiome research methods have been developed in the laboratory of intestinal infections under the guidance of Doctor of Medical Sciences T.P. Haharevaand introduced into the laboratory service of practical health care. The study of the structure of the microbiocrnosis has led to the creation of a system of regenerative therapy for gastrointestinal disorders with using new bacterial preparations of live bacteria - representatives of normal microflora, such as "Colibacterin", "Bifidumbacterin", "Lactobacterin" (the latter for the first time in the country was developed by the staff of the institute and enterprise for the production of bacterial preparations, Blokhina, I. N., Tarasova N. B., Sokolova K. Ya., AkimichevaE. A., Solovyova I. V., etc.).
Currently, an experimental production line of probiotics in the form of liquid biological concentrates based on protein hydrolysate containing live lacto- and bifidumbacteria such as "LB-complex", "LB-complex S", "LL-complex", "LB-complex L" ( Solovyova, I.V., Belova, I.V., Tochilina, AG, Ivanova, T.P., Zhirnov, V.A.) is created.
Since 2010, the laboratory has been headed by Doctor of Biological sciences, Associate Professor I. V. Solovyova.
Directions of activities:
- Study of the microbiota of healthy people and people with various pathological conditions.
- Identification of microorganisms using a new high-tech equipment - MALDI TOF mass spectrometerAutoflex speed (Bruker Daltonik) and software-hardware complexBiotyper.
- Biotechnological developments in the design of new liquid probiotics, the creation of algorithms for their use in a complex of health and medical rehabilitation activities in patients with various nosological forms of diseases complicated by gastrointestinal dysbiosis and their introduction into the therapeutic and preventive work of practical health care organizations.
Laboratory achievements:
- Lactobacillus collection has been created and is being maintained
- Production line of probiotics in the form of liquid biological concentrateshas been created.
- In 2013, patent No. 2441907 for the “Method for preparation of therapeutic product out of living microbial strains of lactic bacteria and bifidobacteriaLB-complex L”developed in the laboratory was awardeda diploma for the 1st place in the nomination "The best invention of the year in the Nizhny Novgorod region in the field of Medicine and Pharmaceutics" and the silver I. Kulibin medal at the 7th competition of intellectual property "Patent of the Year".
- Newly developed probiotics have been successfully used in 22 medical and preventive institutions of Nizhny Novgorod, as well as in the framework of the program for the prevention and rehabilitation of children in 14 preschool educational institutions and in one of the Nizhny Novgorod universities.
Prospects:
- Study and monitoring of antimicrobial resistance of microorganisms: determination of genetic determinants in endemically significant strains of bacteria
- Study of bacterial genes for the presence of single nucleotide polymorphisms (SNP) using MALDI TOF mass spectrometer Autoflex speed and software GenoToolsto identify mutations in the extended-spectrum beta-lactamase genes.
- Molecular proteomics; bacterial cell protein sequencing, study of bacterio-cins of protein nature; analysis of post-translational changes in proteins; study and search for protein markers of bacterial cultures using MALDI TOF mass spectrometer Autoflex speed and softwareBioTools for the purpose of in-depth study of production-promising strains of lactobacilli.
- Creation of prototypes of new probiotics in the form of dietary supplements.
